'Simple and effective' injection could offer hope for treatment of autoimmune disease

Australian researchers have uncovered a potential new way to regulate the body's natural immune response, offering hope of a simple and effective new treatment for auto-immune diseases.

Auto-immune diseases result from an overactive immune response that causes the body to attack itself.

The new approach involves increasing good regulating cells in the body, unlike most current research which focuses on stopping "bad" or "effector" cells, says lead researcher Dr Suzanne Hodgkinson, from UNSW's Faculty of Medicine and Liverpool Hospital.

The researchers induced the body's T-cell front-line defences by injecting cell-signalling proteins called cytokines, in particular cytokine Interleukin-5 (II-5 cytokine).

When T-regulatory cells are grown in a way to make them specific to a particular protein they develop receptors for the Il-5 cytokine. The Il-5 cytokine boost allows the body's immune system to better regulate its response to disease without going into overdrive.

The team cloned II-5 cytokine and injected it into rats with the neurological condition Guillain?Barr? syndrome. These rats recovered much quicker and, if treated as a precaution, did not fall ill. The method has also shown promise in animals with multiple sclerosis, with kidney disease nephritis and trying to overcome organ transplantation rejection.

"One of the nice things about this discovery is that it is one of the few treatments in the auto-immune world and in the transplantation world that works not by attacking the effector cells, but by increasing the good regulating cells. So it works in a very different way from almost every other treatment we've got available," Dr Hodgkinson says.

Il-5 injections could be more palatable than inoculation by parasitic worms ? another approach in regulating auto-immune conditions, the researchers say.

International research shows swallowing helminths parasites can regulate the immune system and boost T-cell production to combat illnesses such as celiac disease and multiple sclerosis. The absence of the worms in guts in the developed world has been cited as a possible cause for the sharp rise in auto-immune diseases in Western nations.

"The process we've developed may be the same process that the helminths kick off. When you get a helminths infestation, one of the changes in your immune response is an increase in cells called eosinophils and these cells make the cytokine Interleukin-5," Dr Hodgkinson says.

"In this new treatment, it's a matter of injecting the interleukin-5 and the body does the rest. It's both safe and effective and we think inducing the immune response by injection may be more attractive to people than swallowing parasitic worms."

The next step is to take the treatment to human trials, which could be underway within two to five years, says Dr Hodgkinson, whose paper outlining the study has been published in the journal Blood.

No charges in Utah 'Sister Wives' case

SALT LAKE CITY (AP) ? A Utah prosecutor says he will not pursue criminal charges against a polygamous family made famous by a reality TV show.

Utah County Attorney Jeff Buhman says he has closed the case against Kody Brown and his four wives, who are the stars of the TLC show "Sister Wives."

Brown moved his family to the Las Vegas area in January 2011 after Utah authorities launched a bigamy investigation.

Brown has since sued Utah County, claiming Utah's bigamy statute violates his family's constitutional rights.

Buhman is now seeking to have the lawsuit dismissed, claiming it is moot given the case is closed.

He says his office has also adopted a policy that bigamy cases will only be pursued if there is evidence of a victim or fraud.

A former Rutgers University student openly apologized for the first time on Tuesday for using a webcam to spy on a romantic liaison between a man and a roommate who later killed himself, saying he regrets his "thoughtless, insensitive, immature, stupid and childish choices."

Dharun Ravi, who had been criticized by a judge for not showing remorse and for refusing to say he was sorry, also said he will begin serving a 30-day jail term on Thursday even though he doesn't have to.

Through a lawyer, Ravi issued his most contrite public statement yet in a case that made him a symbol of what his family called an overzealous prosecution and that made his roommate, Tyler Clementi, a prime example of what gay rights advocates said were the consequences of bullying.

"I accept responsibility for and regret my thoughtless, insensitive, immature, stupid and childish choices that I made on Sept. 19, 2010, and Sept. 21, 2010," Ravi said in his statement. "My behavior and actions, which at no time were motivated by hate, bigotry, prejudice or desire to hurt, humiliate or embarrass anyone, were nonetheless the wrong choices and decisions. I apologize to everyone affected by those choices."

After spending two days repeatedly looking at the Twitter feed on which Ravi announced "I saw him making out with a dude. Yay," Clementi, a shy, talented violinist, threw himself from New York City's George Washington Bridge on Sept. 22, 2010.

In March, a jury convicted Ravi of all 15 criminal counts with which he was charged, including invasion of privacy and bias intimidation. On two of the intimidation counts, he faced up to 10 years in state prison.

Last week, a judge sentenced him to 30 days in jail. Because the sentence is less than a year, it decreases the chances that federal immigration authorities will seek to have Ravi deported to India, where he was born and remains a citizen.

Prosecutors, finding the sentence too lenient, said they would appeal.

Ravi's lawyers have said they expect to appeal the convictions entirely. They say that he was not hateful and that authorities charged him with such serious crimes because of Clementi's suicide even though he was not charged with the 18-year-old's death.

Ravi, 20, could have remained free during the appeal but instead is volunteering to head to the Middlesex County Jail in New Brunswick.

"It's the only way I can go on with my life," he said in the statement.

The apology comes as a sharp reversal in course for Ravi, whose story inspired hundreds of people to rally at New Jersey's State House calling for no prison time and changes in the state's hate crime laws.

When Ravi was sentenced, Judge Glenn Berman chastised him for not apologizing for his actions.

"I heard this jury say 'guilty' 288 times," the judge said, referring to all the sub-parts of the charges Ravi faced repeated 12 times, once for each juror. "And I haven't heard you apologize once."

During the court proceeding, Ravi, who had said in March in a newspaper interview that he was "very sorry about Tyler," chose not to address the judge, though he cried as his mother pleaded for mercy for him.

He told Newark's The Star-Ledger newspaper in an interview conducted before the sentencing but published afterward that he did not want to say he was sorry during the sentencing because he thought it would sound insincere.

During the sentencing, Clementi's brother James Clementi said that hearing an apology that late from Ravi would not be meaningful to him.

On Tuesday, the state's largest gay rights group, Garden State Equality, said it was happy Ravi had publicly apologized. But Chairman Steven Goldstein said the group was questioning the timing of the apology.

"We have mixed emotions, and so rather than take an organizational stance just yet, we have posited the question to our members on (the group's) Facebook page to ask what they think," Goldstein said.

Garden State Equality has said Ravi deserves more jail time than he received but "nowhere near" the maximum sentence he could have received.

"We have said that our hearts would be open to an apology had Ravi opened his heart up to us all," Goldstein said. "He could have apologized in a way that would not have compromised his legal position. But he came across to many of us as unremorseful both before the trial and in engineered media appearances after the verdict."

Tinley mental health center closing plan needs ... - Health and Fitness

Just five weeks before the scheduled closing of the Tinley Park Mental Health Center, service providers and advocates say the transition plan comes with too little funding to safely serve residents of the south suburbs.

Gov. Pat Quinn announced the closing this year as part of a ?rebalancing plan? that will transition those with mental illnesses and developmental disabilities out of institutions and into community-based settings. The center has also been lumped into the governor?s cost-cutting plan to close and consolidate state facilities.

Despite a legislative panel?s recommendation to keep the center open, Quinn has pushed forward with the plan.

Department of Human Services officials last week reviewed the closing plan with the providers that will replace services offered by the center. Lorrie Rickman Jones, director of the department?s Division of Mental Health, said DHS is ready to shut down the 54-year-old facility by July 1, despite recent legislative attempts to keep it open for another six months to allow for more planning.

?We?re ready to go,? she said. ?The contracts are on the street and the money will be on the street very shortly.?

DHS accepted proposals from 10 hospitals, seven community health providers and four substance-abuse service providers for replacement services once the center closes, she said, but contract negotiations are ongoing.

Mental health advocates and lawmakers alike have criticized DHS for having too few details settled for serving residents when Tinley?s center closes. With the last date for patient admission set for June 15, stakeholders say they are happy for more details but still have concerns. Chief among them is that the plan doesn?t offer enough funding for the services that hospitals and community providers are taking on.

?Everyone shares the concerns around sustainability and whether there?s enough funds to take care of the transition,? said Frank Anselmo, CEO of Community Behavioral Healthcare Association of Illinois.

Jones said DHS will spend about $10 million on the plan, which is estimated to serve 1,400 clients a year through inpatient, outpatient and substance-abuse services. With an operating budget of $20.3 million in fiscal year 2011, the facility served 1,900 patients.

Anselmo said that doesn?t account for others who never got treatment because of a shortage of psychiatric beds for the uninsured in the Chicago area.

DHS said the new plan is designed to redirect some patients out of hospital beds and into substance-abuse treatment or outpatient care programs.

?Our studies estimate that we will see the need for admissions decrease by about 45 percent,? DHS spokeswoman Januari Smith Trader said in an email.

Joseph Troiani, director of behavioral health programs for the Will County Health Department, said about half the patients sent to the Tinley Park center through his programs have a substance-abuse problem.

Troiani said the new system of assessing patients is a plus. Though he described the overall plan as ?workable,? he added that it would benefit from additional resources.

Jackie Haas, president and CEO of The Helen Wheeler Center, a mental health provider in Kankakee, said she has yet to sign a contract with DHS and is moving forward cautiously because of funding concerns.

Ingalls Memorial Hospital in Harvey is still reviewing a contract to provide psychiatric beds for DHS, said Jeff Bergren, director of behavioral health. The hospital referred 220 patients to Tinley Park last year, he said, but the state contract would cover only 60 percent of those patients.

?It would seem to say we are not going to have sufficient funding to take care of these patients throughout the year,? he said. ?I think DHS has actually done a pretty good job of putting together a system of care with the available resources they have, but I don?t think the available resources are sufficient.?

'Fair' pricing? Hell no, we like being shafted!

You might have seen recently that iconic retailer JC Penney is slumping badly. You almost certainly have seen the reason why: A massive, creative and aggressive new advertising and pricing campaign that promises simplified prices.

No more coupons or confusing multiple markdowns. No more 600 sales a year. No more deceptive circulars full of sneaky fine print.?Heck, the store even did away with the 99 cents on the end of most price tags.? Just honest, clear prices.

Sounds like a sales pitch aimed at consumer advocates and collectors of fine print frustration, like me. As it turned out, it was a sales pitch that only a consumer advocate could love.

Shoppers hated it.

The campaign, which launched on Feb. 1, appears to be a disaster. Revenue dropped 20 percent for the first quarter compared to last year. Customer traffic fell 10 percent. Last year, the company made $64 million in the first quarter; this year, it lost $163 million.

Not only did Penney?s plain pricing structure fail to attract fair-minded shoppers ? ?business reporters wrote with seeming glee during the past few days that it??repelled? them.

Don't blame Ellen DeGeneres, the spokeswoman for the Penney?s plain pricing campaign. If only executives at the firm were familiar with the work of behavioral economist Xavier Gabaix and the concept of "shrouding," all of this could have been avoided.

Seven years ago, Gabaix and co-author David Laibson wrote a brilliant (if depressing) paper on shrouding and "information suppression" that should be required reading for all consumers and executives considering a harebrained new pricing strategy.?The principle is simple, and shows why cheating is rampant in our markets and why honesty is rarely the best policy.

First, a definition of shrouding:

In days gone by, price tags were simple. An apple cost 10 cents.? A cup of coffee cost $1.?But today, the consumer marketplace is far more complicated, giving sellers the opportunity to create confusion. Many items have follow-up costs that make the original price tag meaningless.?

Computer printers are the classic example. You might get a great deal on a printer, but if the ink is expensive, you lose in the end. In fact, Gabaix argues that it's impossible for consumers to intelligently shop for printers. No consumer knows how much ink costs -- the cartridges don't come in standard sizes, the amount of ink used to print varies and ink costs are unpredictable.?That makes the true price of a printer "shrouded," in Gabaix's terminology. Not quite hidden, but not quite clear, either.? Advantage seller.?It's easy for printer companies to lowball printer price tags and overcharge for ink, enabling them to print money.

If you think about it, shrouded price tags are everywhere.?The hotel website might say "$99 a night" but you know the bill will be more like $120 or $130. Pay TV companies promise $30-a-month service, which ends up costing more like $50.?And what happens when you buy a TV with a store credit card that offers an upfront discount but a complex interest charge? And so it goes.

Consumers complain about this constantly.?That's the basis of the Red Tape Chronicles in fact. At its best, the maddening mixture of coupons, rebates, sales and fine print fees can feel like a game. At worst, it's being cheated.?You'd think shoppers would love a chance to buy from a store that doesn't play these games, the way car buyers (allegedly) like shopping at no-haggle auto dealerships.

They don?t, says Gabaix, and Penney should have known better.

?I think it was an ill-advised move,? he said.?

All this price manipulation is really an information war, he says.?Shoppers hunt for the tricks that let them save money.?Stores hide booby traps that let them take money.?It's a bad system, one I've labeled "Gotcha Capitalism." But it is the system we have now.

And it's simply impossible, Gabaix argues, to be the one company that attempts to bridge this information gap.? If a firm tries to educate consumers on tricks and traps, and tries to offer an honest product, a funny thing happens: Consumers say, "Thank you for the tips," and go back to the tricky companies, where they exploit the new knowledge to get cheaper prices, leaving the "honest" firm in the dust.

?Once you educate consumers on the right way to shop, they will seek out the lowest cost store, and that will be the one with the shrouded prices,? he said. ?Once they are savvier consumers, you make less money from them.?

Gabaix calls this the "curse of debiasing." And it leads to this depressing conclusion: "Shrouding is the more profitable strategy."

To oversimplify for a moment, here's Penney's problem.?They told the world that retailers only offer their best prices during crazy sales, and Penney stores would no longer host them.?Sensible consumers apparently took that information to heart and decided to simply wait for such sales at other stores. As an added benefit, Penney lowered consumers' search costs, because they now knew they didn't need to bother driving to a Penney?s store anymore.

That's probably not what new Penney CEO Ron Johnson had in mind when he decided to spend his marketing budget on those witty DeGeneres ads. A former Apple Inc. executive who took the Penney?s job in November, he thought he was lifting the store out of the brutal commodity clothing market. He may ultimately succeed at that. But he won't do it by telling customers the firm's pricing is fairer than at other stores, Gabaix believes.

"It will be a very, very uphill battle," Gabaix said. "So, sorry for them."

There have been a few other celebrated efforts by companies to educate consumers that their higher prices are really lower prices after hidden fees.?During the last decade, Intercontinental Hotels experimented with up-front pricing that included all fees on its website. Executives at the firm told the New York Times that customers left in droves, choosing competitors with lowball prices.?

More recently, Southwest Airlines has undertaken the most aggressive anti-shrouding campaign to date, picking on other airlines' baggage fees. The profitable carrier is holding its own with its "Bags Fly Free" campaign, but there are indications that the firm won't be able to resist all that free money forever. In what may be a sign of things to come, Southwest elected to leave AirTran's baggage fee structure in place after it acquired the competitor last year.?

Shrouding isn't the only reason Penney's pricing plan is flawed.?The firm is also leaving a lot of money on the table by rejecting a phenomenon known as "price discrimination."?Some people have more money than time, and some have more time than money.? Some shoppers don't mind spending hours to save $20; others would gladly give a store $20 to escape quickly.?Smart retailers get money from both.?By killing couponing, Penney has eliminated its ability to satisfy price discriminators.

And as others have pointed out, markdowns serve the age-old retailing trick of "anchoring." For some reason, even very smart consumers feel better paying $60 for something if you initially tell them it costs $100, and then reduce the price.

But the real problem is Penney's ill-fated attempt to cast itself as the only fair poker player in a game of cheats.?Shoppers just aren't buying it. However unsophisticated consumers are, very few of them believe a pair of shoes bought at Penney's everyday low price will be cheaper than a pair of shoes bought at Macy's on clearance with a 25 percent off coupon.

Like it or not, hidden fees ? and secret discounts ? are here to stay.

PAN.THY.MONIUM Khaoohs music review by sagichim

Pan.Thy.Monium's second album Khaoohs is pretty much a continuation from where they left us with Dawn Of Dreams. For you who are unaware of this group, it's another project out of the many headed by Dan Swano, well actually it's not just another project, it's the most progressive one and maybe the heaviest one by him, and if i may say, it's definitely my favorite. Swano uses this band to go wild, to be weird and most of all to experiment not just with music but on the vocals department too. Although known for his great vocals, clean or growls, Swano leaves the growling job for his band mate Robert Karlsson, which is doing a great job and manages when he likes, not to be harsh but as pleasant as those vocals can be. The music essentialy is a good mix between extreme metal, jazz and other avant ideas. I'm not a fan of the extreme metal genre at all, but this group just grabbed me with their cool mix of styles and above all great playing.

What makes them one of the best progressive acts are two things, one is their instrumentation, that aside from your usual metal music instruments there's the addition of keys and saxophone, the second is the fact that they are totally progressive, and knows how to write good effective music, the ideas are unusual and interesting, the music is going forward and is not being stuck on a main riff idea, it spreads to all kinds of directions. The band is tight and just knows how to play together, it all sounds very coherent and powerful. Another element that characterizes the band are the guitars, rhythm guitar has a very low sludgy tone that suits the vocals perfectly since they are low and sludgy too, Lead guitar does not have that typical extreme metal tone or that metal way of playing, the sound is very clear and not that distorted, and has very jazzy touch. The solos are amazing, played a lot of times with wah wah which makes it very groovy, with all sorts of jazzy lines and some bluesy lines too. Swano's bass playing is also good as he uses some chorus effect at times, giving the music some melodic touch.

Although half the album contains short songs, that doesn't hold them of being any less progressive than the longer cuts, every song is going in it's own progy way, the ideas are not repeating theirselves, and every song contain something unique, like saxophone/guitar riffs, funky jazzy solos, beautiful melodic parts, all sorts of keys parts, avant-garde and excellent riff after riff. The music goes from slow doomy riffs to faster breaks and all sorts of avant progressive additions. I believe nothing can prepare you to what you are about to hear because it's all pretty unexpected. I wasn't impressed the first time i heard it but definitely felt something special. As it grew on me every time i played it, i realize that it might be better than their debut. You can't go wrong with any of their albums, and this one is also superb. 4.2 stars for this genre. Recommended!!
